După despărțirea de fostele colege O-Ren Ishii (Lucy Liu) și Vernita Green (Vivica A. Fox) în Kill Bill: Vol. 1, Mireasa (Uma Thurman) își reia aventura pentru dreptate în a doua parte. Învingându-le pe cele două, Mireasa mai are de găsit doi dușmani de pe "Lista Morții" - Budd (Michael Madsen) și Elle Driver (Daryl Hannah) - înainte de a se îndrepta spre scopul ultim: uciderea lui Bill (David Carradine).
